چکیده

Secretion occurs in all cells of multicellular organisms and involves the delivery of secretory products packaged in membrane-bound vesicles to the cell exterior. Specialized cells for neurotransmission, enzyme secretion or hormone release utilize a highly regulated secretory process. Secretory vesicles are transported to specific sites at the plasma membrane, where they dock and fuse to release their contents. Similar to other cellular processes, cell secretion is found to be highly regulated and a precisely orchestrated event. It has been demonstrated that membrane-bound secretory vesicles dock and fuse at porosomes, which are specialized supramolecular structures at the cell plasma membrane. Swelling of secretory vesicles results in a build-up of pressure, allowing expulsion of intravesicular contents. The extent of secretory vesicle swelling dictates the amount of intravesicular contents expelled during secretion. The discovery of the porosome, its isolation, its structure and dynamics at nm resolution and in real time, its biochemical composition and functional reconstitution into artificial lipid membrane, have been determined. The molecular mechanism of secretory vesicle fusion at the base of porosomes, and vesicle swelling, has also been resolved. These findings reveal the molecular mechanism of cell secretion.

During the last five years the recognition of ionic channels in the parietal cells of stomach and acid chloride mechanisms of secretion by these cells has become totally clear by the "Patch Oamp" technique. The apical cytoplasm in the oxyntic cells are in the form of vesicles where membranes contain H+, K+ -ATPase pump. Stimulation causes fusion of these tubular vesicles with the cell membran o...

O-8: Molecular Mechanisms of Membrane Fusion Involved in Fertilization

Background: Assisted fertilization procedures are a currently widespread practice to regulate reproduction in humans and animals. The arising question is why the human being manipulating gametes to generate new individuals, if we do not understand yet the molecular mechanism of fertilization?. Successful completion of fertilization in mammals is dependent on three membrane fusion events: 1. the...

Membrane fusion/repair in nerve cells: a biophysical application in spinal cord injuries regeneration

Cell membrane has a critical and vital role in functioning and existence of nerve cells that form central nervous system (CNS) in mammals. Disruption of nerve membrane that normally occurs following an accident injuring spinal cord is known to be the major cause of paralysis. In most occasions, spinal cord injuries are not leading to complete cut in spinal cord fibers but are known to cause cru...

Secretion is required for late events in the cell-fusion pathway of mating yeast.

Secretory vesicles accumulate adjacent to the contact site between the two cells of a yeast mating pair before they fuse, but there is no direct evidence that secretion is required to complete fusion. In this study, temperature-sensitive secretion (sec(ts)) mutants were used to investigate the role of secretion in yeast cell fusion. Cell fusion arrested less than 5 minutes after inhibiting secr...
